Intended Use
Surra is a severe, economically devastating parasitic disease affecting a wide range of domestic and wild mammals, most notably camels, horses, cattle, water buffaloes, and dogs. The disease is caused by Trypanosoma evansi, a pathogenic flagellated protozoan parasite. Unlike other trypanosomes, T. evansi is mechanically transmitted via the bites of hematophagous biting flies. As a consequence, surra is recognized as a high-consequence transboundary disease by the WOAH due to its wide geographical distribution across Asia, Africa, and Latin America, and its severe impact on livestock productivity and draft animal welfare. The «Trypanosoma evansi qPCR kit» is intended to detect Trypanosoma evansi DNA in whole blood, serum, plasma, lymph node aspirates, and animal tissue samples, providing a rapid, accurate, and easy-to-operate diagnostic solution. The test kit «Trypanosoma evansi qPCR kit» is designed for multiplexed detection of DNA specific to Trypanosoma evansi (targeting the highly conserved ITS rRNA gene) and an Internal Control as a sample processing control by realtime polymerase chain reaction (PCR) with fluorescent detection of the analysis results.
Principle

The method is based on the isolation of total DNA from samples, and further amplification of specific regions of T. evansi (ITS gene) using specific oligonucleotide primers and the synthesis of complementary DNA chains using the enzyme Hot start Taq-DNA polymerase. Fluorescent detection of the analysis results is performed both in real-time (Real-time mode) or end point assay using two fluorescence detection channels (FAM525 nm, ROX580 nm).

Blood, tissue, swabs samples can be used directly for Nucleic Acids extraction according commercially available DNA extraction kit recommendations.
