Intended Use
Lumpy skin disease (LSD) is a poxvirus disease of cattle characterized by fever, nodules on the skin, mucous membranes and internal organs, emaciation, enlarged lymph nodes, oedema of the skin, and sometimes death. The «LSDV qPCR kit» is intended to detect viral DNA in blood, animal tissue, swabs semen and environmental samples, which is rapid, accurate and easy-to-operate. The test kit «LSD qPCR kit» is designed for multiplexed detection of the specific to LSDV DNA (PolyA gene), according OIE validated protocol, and Internal Control (Ruminant gene) as sample processing control by real time polymerase chain reaction (PCR) with fluorescent detection of the analysis results.
Principle

The method is based on the isolation of total DNA from samples, and further amplification of specific regions of LSDV (PolyA gene) using specific oligonucleotide primers and the synthesis of complementary DNA chains using the enzyme Hot start Taq-DNA polymerase. Fluorescent detection of the analysis results is performed both in real-time (Real-time mode) or end point assay using two fluorescence detection channels (FAM525 nm, ROX580 nm).

Blood, tissue, swabs and semen samples can be used directly for Nucleic Acids extraction according commercially available DNA extraction kit recommendations.
