Intended Use
Glanders is a contagious and fatal disease of horses, donkeys, and mules, caused by infection with the bacterium Burkholderia mallei. The pathogen causes nodules and ulcerations in the upper respiratory tract and lungs. The «Glanders qPCR kit» is intended to detect Burkholderia mallei/pseudomallei DNA in blood, tissues, swabs, feces and cultural material, which is rapid, accurate and easy-tooperate. The test kit «Glanders qPCR kit» is designed for multiplexed detection of the specific to Burkholderia mallei/pseudomallei DNA (aroA gene), according OIE validated protocol, and Internal Control (16 S) gene as sample processing control by real time polymerase chain reaction (PCR) with fluorescent detection of the analysis results
Principle
The method is based on the isolation of total DNA from samples, and further amplification of specific regions of Burkholderia mallei/pseudomallei DNA (aroA gene) using specific oligonucleotide primers and the synthesis of complementary DNA chains using the enzyme Hot start Taq DNA polymerase. Fluorescent detection of the analysis results is performed both in real-time (Real-time mode) or end point assay using two fluorescence detection channels (FAM525 nm, ROX580 nm).
Blood, tissue, swabs and semen samples can be used directly for Nucleic Acids extraction according commercially available DNA extraction kit recommendations.
